A leaf, leaflet, or leaf portion was removed, cytoplasm
cleared by treatment with ethanol and Clorox™, dehydrated
with ethanol, permanently cleared with xylol, and mounted
in Permount to visualize calcium oxalate crystals in the
lamina. Cleared lamina was viewed between crossed
polarizers using an LM to determine crystal shape and
arrangement, as part of a larger project to compare leaf
crystal patterns within the Glycininae to previously
published molecular data.
